Students will use the supplies from this project to do an advanced biotechnology technique called SDS-PAGE which uses a gel to separate proteins by size. In this lesson, students are developing and using biotechnology skills to determine the evolutionary relationship of a set of fish. I also include a "mystery fish" which students need to place-based only on its banding pattern. Students will pipette the fish protein into gels, separate the proteins by size using electrophoresis, and then examine the banding patterns in the stained gels to determine which species are the most related. This is an amazing lab that teaches many practical biotechnology skills as well as key biology concepts, but the materials are expensive enough that most students are never able to experience them. Because I have scaffolded the lesson to meet my students where they are, my entry-level Biology students have been able to successfully complete this rigorous lab. Their success has inspired them to feel more confident in their abilities and to at least consider continuing in upper-division science classes.
